作者对置液氮中冻存八年复苏后的个旧肺腺癌细胞系(GLC-82)的生物学特性进行观察。结果:复苏癌细胞经台盼兰染色在光镜下观察90%以上为活细胞。H·E染色后在光镜下见癌细胞胞膜完整,呈多形性,核仁明显。细胞复苏后培养倍增时间为46小时,与冻存前基本相似。液氮冻存前染色体众数为63-65,复苏培养后测定为62-65,性染色体可明显辨认,配对组型没有明显改变。在透视电镜下部分细胞浆内查见成熟程度不同的板层小体,有少量粗面内
The authors examined the biological characteristics of Gejiu lung adenocarcinoma cell line (GLC-82) after cryopreservation in liquid nitrogen for eight years. Results: More than 90% of the resuscitated cancer cells were observed by trypan blue staining under light microscope. H · E staining in the light microscope, see the cell membrane integrity, was pleomorphic, obvious nucleoli. Culturing doubling time after cell resuscitation was 46 hours, basically similar to that before cryopreservation. The number of chromosomes before liquid cryopreservation was 63-65, and the number of chromosomes after the resuscitation was 62-65. The sex chromosomes were clearly identifiable and the paired group did not change significantly.
