目的采用高效毛细管电泳法,建立宁夏枸杞中N-反式阿魏酸酪酰胺含量测定方法。方法未涂层毛细管柱(50 cm×75μm,有效长度为40 cm),以50 mmol.L-1硼砂溶液(pH 9.0,磷酸调节)为运行缓冲液,运行电压为20 kV,柱温25℃,检测波长214 nm。结果在进样量0.25~15μg.mL-1内线性关系良好,相关系数r=0.999 8(n=7)。N-反式阿魏酸酪酰胺的平均回收率为101.8%,RSD为2.06%。结论该方法准确可靠,可用于测定宁夏枸杞中N-反式阿魏酸酪酰胺的含量,为宁夏枸杞开发利用提供理论依据。
Objective To establish a method for the determination of N-trans-ferulic acid tyrosamide in Lycium barbarum L. by capillary electrophoresis. Methods Uncoated capillary column (50 cm × 75 μm, effective length 40 cm) was applied with 50 mmol·L-1 borax solution (pH 9.0, adjusted with phosphoric acid) as the running buffer. The operating voltage was 20 kV and the column temperature was 25 ℃ , Detection wavelength 214 nm. The results showed good linearity in the range of 0.25 ~ 15μg.mL-1, the correlation coefficient was 0.999 8 (n = 7). The average recovery of N-trans-ferulic acid amide was 101.8% with a RSD of 2.06%. Conclusion The method is accurate and reliable and can be used to determine the content of N-trans-ferulic acid amide in Lycium barbarum, providing a theoretical basis for the development and utilization of Lycium barbarum in Ningxia.
