目的:建立荷叶碱(NF)、氮-去甲荷叶碱(N-NF)和2-羟基-1-甲氧基阿朴啡(HMA)的LC-MS/MS分析方法,对其在大鼠的组织分布进行研究.方法:通过给大鼠静脉注射荷叶总生物碱10 mg·kg-1后,采用LC-MS/MS方法测定各组织中主要生物碱成分NF,N-NF和HMA的含量.结果:给大鼠静脉注射总生物碱后,3个主要生物碱成分NF,N-NF和HMA能迅速(5 min)分布到各组织,其中在肝脏、肺、脾脏和肾脏等组织中具有较高的浓度,6h后显著下降.结论:建立的方法简便易行、准确可靠,可用于大鼠体内NF,N-NF和HMA的组织分布研究;NF,N-NF和HMA在大鼠体内分布快且广泛,并在体内维持较长时间.“,”Objective:To establish an LC-MS/MS method for the analysis of tissue distribution of nuciferine (NF),N-nornuciferine (N-NF) and 2-hydroxy-1-methoxyaporphine (HMA) in rats.Methods:Total alkaloids were administrated intravenously (10 mg· kg-1) to rats and the concentrations of NF,N-NF,and HMA in biological samples were determined with established LC-MS/MS method.Results:The results showed that NF,N-NF and HMA could be quickly (within 5 min) distributed to various tissues,especially in the liver,lung,spleen and kidney with relative high concentrations,and decreased significantly in 6 hours in most tissues.Conclusion:The established method is simple and accurate,and can be used to study the tissue distribution of NF,N-NF and HMA in rats.The NF,N-NF and HMA are widely distributed and remained in the bodies for a relative long period.
