【目的】探讨蒲黄总黄酮(PTF)对饱和脂肪酸棕榈酸(PA)诱导损伤INS-1胰岛β细胞的影响。【方法】采用长期PA诱导INS-1胰岛β细胞的方法建立细胞损伤模型,并给予PTF干预处理,采用二甲氧唑黄(XTT)比色法检测细胞活性。【结果】PA呈浓度和时间依赖性损伤INS-1胰岛β细胞活性,而PTF则呈浓度依赖性改善PA对INS-1胰岛β细胞的损伤。此外,PA对INS-1胰岛β细胞损伤的幅度越大,PTF对损伤的改善作用也就越大。【结论】PTF能改善PA长期诱导所致的INS-1胰岛β细胞损伤。
【Objective】 To investigate the effect of total flavonoids (PTF) from Pueraria purpurea on the injury of INS-1 β-cells induced by saturated fatty acid palmitate (PA). 【Methods】 Long-term PA induced INS-1 islet β cells were used to establish a cell injury model, and PTF intervention was performed. Cell viability was measured by XTT colorimetry. 【Results】 The results showed that PA damages INS-1 islet β-cell activity in a concentration-and time-dependent manner, while PTF exerts a concentration-dependent improvement in PA-induced damage to INS-1 β-cells. In addition, the greater the extent of PA damage to INS-1 islet β cells, the greater the effect of PTF on the damage. [Conclusion] PTF can improve the long-term induction of PA induced INS-1 islet β cell injury.
