目的研究人和小鼠白血病细胞膜流动性(MF)的变化及卟啉胆固醇酯对人早幼粒细胞性白血病细胞(HL┐60细胞)和人霍奇金淋巴瘤细胞(Daudi细胞)MF的影响动态观察二甲基亚砜(DMSO)诱导后的HL┐60细胞MF的改变。方法以DPH为荧光探针,用荧光偏振技术检测MF。结果Daudi细胞较健康人外周血淋巴细胞膜流动度增高1.6倍;白血病小鼠脾淋巴细胞,培养的小鼠Ts淋巴白血病细胞较正常小鼠脾淋巴细胞及T淋巴细胞MF明显增高(P<0.001);卟啉胆固醇能明显降低HL┐60细胞和Dauid细胞MF,二甲基亚砜(DMSO)诱导后的HL┐60细胞MF降低30%(P<0.01)。结论MF的增加确实是某些肿瘤细胞的重要特征之一,但这种特征可随着药物治疗及癌细胞分化成熟而逆转。
Objective To investigate the changes of membrane fluidity (MF) in human and mouse leukemia cells and the effect of porphyrin-cholesteryl ester on MF in human promyelocytic leukemia cells (HL┐60 cells) and human Hodgkin’s lymphoma cells (Daudi cells). The changes of MF in HL┐60 cells induced by dimethyl sulfoxide (DMSO) were observed dynamically. Methods DPH was used as fluorescent probe and MF was detected by fluorescence polarization technique. Results Membrane fluidity of peripheral blood lymphocytes in Daudi cells was 1.6-fold higher than that in healthy individuals. Spleen lymphocytes and cultured mouse Ts lymphocytic leukemia cells were significantly higher than those of normal mouse splenic lymphocytes and T lymphocytes (P< 0.001); porphyrin cholesterol can significantly reduce HL ┐ 60 cells and Dauid cells MF, DMSO-induced HL ┐ 60 cells MF decreased by 30% (P lt; 0.01). Conclusion The increase of MF is indeed one of the important features of certain tumor cells, but this feature can be reversed with drug treatment and differentiation and maturation of cancer cells.
