应用疏水层析法纯化hCG单克隆抗体

建立一种良好的纯化hCG单克隆抗体方法。将hCG单克隆抗体的实验小鼠腹水经离心、过滤预处理后,用疏水层析法进行纯化,并探讨了洗脱方式、不同上样缓冲液离子强度、洗脱体积对抗体纯度的影响。以20 mmol/mL磷酸缓冲液+1.5 mmol/L硫酸铵为上样缓冲液,15 CV的洗脱体积进行洗脱,单抗纯度为70%,回收率20%。纯化后的单克隆抗体的生物学活性没有下降。本研究建立的疏水层析法纯化hCG单克隆抗体的方法,操作简便、快速而且效果良好。 Establish a good method of purification of hCG monoclonal antibody. The ascitic fluid of mouse hCG monoclonal antibody was centrifuged, pretreated by filtration and purified by hydrophobic chromatography. The effects of the elution mode, the ionic strength of different loading buffer and the purity of antibody on elution volume were discussed. Elution was performed with 20 mmol / mL phosphate buffer + 1.5 mmol / L ammonium sulfate as loading buffer and 15 CV elution volume. The purity of monoclonal antibody was 70% and the recovery rate was 20%. The biological activity of the purified monoclonal antibody did not decrease. The method of hydrophobic chromatography to purify hCG monoclonal antibody established in this study is simple, rapid and effective.
