目的观察转染BMP2和VEGF165双基因共表达质粒在小鼠骨髓基质干细胞的表达情况。方法脂质体介导下将双基因真核表达质粒pIRES-BMP2-VEGF165导入小鼠骨髓基质干细胞,用RT-PCR和免疫组织化学方法观察BMP2和VEGF165双基因在小鼠骨髓基质干细胞内的表达。结果转染BMP2、VEGF165的小鼠骨髓基质干细胞有明显的BMP2和VEGF165mRNA及其蛋白表达。结论转染BMP2和VEGF165双基因的小鼠骨,髓基质干细胞能同时表达以上两种基因。
Objective To observe the expression of BMP2 and VEGF165 double gene co-expression plasmid in mouse bone marrow stromal cells. Methods Liposome-mediated eukaryotic expression plasmid pIRES-BMP2-VEGF165 was transfected into mouse bone marrow stromal cells. The expression of BMP2 and VEGF165 double gene in mouse bone marrow stromal cells was observed by RT-PCR and immunohistochemistry . Results BMP2 and VEGF165 transfected mouse bone marrow stromal stem cells showed significant BMP2 and VEGF165 mRNA and protein expression. Conclusion Both mouse bone marrow-derived mesenchymal stem cells transfected with BMP2 and VEGF165 genes can express these two genes simultaneously.
