基因治疗 1型糖尿病是近几年的研究热点。将分泌胰岛素的装置导入患者自身的非β细胞 ,使之分泌胰岛素 ,又不受自身免疫系统的攻击。转基因治疗必须具备以下几个方面 :有效的胰岛素基因转载系统 ;准确调节胰岛素分泌的系统 ;胰岛素的加工装置及合适的宿主细胞。其中 ,重组腺相关病毒是最常用的载体 ,K细胞是最理想的宿主细胞。胰岛素基因治疗的主要问题是非 β细胞中缺乏一个能根据血糖水平准确调节胰岛素合成及分泌的系统。随着人们在胰岛素基因治疗方面取得的进展 ,根治糖尿病将成为可能。 Gene therapy type 1 diabetes is a hot research topic in recent years. The insulin-secreting device is introduced into the patient’s own non-beta cells to secrete insulin and is not attacked by the autoimmune system. Transgenic treatment must have the following aspects: a valid insulin gene transfer system; accurate regulation of insulin secretion system; insulin processing device and the appropriate host cells. Among them, recombinant adeno-associated virus is the most commonly used vector, K cells are the most ideal host cells. The main problem with insulin gene therapy is the lack of a system in non-beta cells that accurately regulates insulin synthesis and secretion on the basis of blood glucose levels. With people in insulin gene therapy progress, cure diabetes will be possible.
