目的构建三叶木通AFLP指纹图谱,为种质资源鉴定、品种选育奠定基础。方法采用扩增酶切片段多态性(AFLP)技术,对10份三叶木通种质进行基因组DNA多态性分析。结果从64对引物中筛选到8对高效扩增引物,获得861个多态性位点,多态性比率达到90.65%。依据多态性位点可将10个三叶木通种质聚类为三类。结论 AFLP技术适于三叶木通种质多态性分析。
Objective To construct AFLP fingerprints of Cloverwood and lay the foundation for identification of germplasm resources and breeding of varieties. Methods Genomic DNA polymorphism analysis was performed on 10 accessions of Clover (Cletora communis L.) through amplified fragment amplified polymorphism (AFLP). Results Eighty-four pairs of primers were screened from 64 pairs of primers and 861 polymorphic loci were obtained. The polymorphism rate was 90.65%. According to the polymorphic loci 10 Clover populations can be clustered into three categories. Conclusion AFLP is suitable for the analysis of the genetic diversity of Clover.
