以丹酚酸B为样品,通过对氮蓝四唑光照法评价体系中测定波长,缓冲液pH值、浓度,光照强度及时间等关键因素进行优化筛选,确定丹酚酸类化合物清除O2-.的最佳评价体系,并对丹参中5种水溶性成分进行活性评价。结果表明,最佳测定条件为:测定波长625 nm,磷酸盐缓冲液pH值7.8,缓冲液浓度5 mmol.L-1,光照强度6 000 lx,光照时间30 min,其可靠性好,清除率测定结果的灵敏度较改进前提高20%;丹参5种水溶性成分清除O2-.的能力均随其浓度增加而逐渐增强,线性关系范围内,清除O2-.能力大小依次为:迷迭香酸>丹酚酸A>丹酚酸B>原儿茶醛>紫草酸≈芦丁。
Taking salvianolic acid B as the sample, we optimized the screening of key factors such as the determination wavelength, the pH value of the buffer, the concentration, the light intensity and the time in the system of light blue light blue tetrazolium to determine the salvianolic acid compound to eliminate O2-. The results showed that the best determination conditions were as follows: the wavelength was 625 nm, the pH value of phosphate buffer was 7.8, the concentration of buffer was 5 mmol.L-1, the light intensity was 6 000 lx and the light time was 30 min. The sensitivity of the determination results was improved by 20% compared with that of the prior art; the ability of 5 water-soluble components of Salvia miltiorrhiza to scavenge O2- increased gradually with the increase of its concentration, and within the linear range, O2- was abolished. > Salvianolic acid A> salvianolic acid B> protocatechuic aldehyde> purple acid ≈ rutin.
