目的:研究杂种犬硬膜外注射吗啡(Mor)后,在血浆和脑脊液(CSF)的药物动力学过程,为临床术后硬膜外注射小剂量Mor镇痛提供可靠的实验依据.方法;取健康杂种犬5只,硬膜外注射0.1mg/kg Mor后,采用HPLC法,检测不同时间的血浆和CSF浓度,经3P87软件拟合,并计算药物动学参数.结果:Mor在血浆及CSF的药物—时间过程符合二室模型,C_(max)分别为28.21、126.31ng/ml(P<0.01).AUC为2174.39、16 650.90ng·min/ml(P<0.01),T_(1/2β)为116.71、330.49min(P<0.05).结论:硬膜外注射Mor,CSF中的药物浓度高.并且维持时间长,有利于长时间镇痛.
OBJECTIVE: To study the pharmacokinetics of morphine (Mor) in plasma and cerebrospinal fluid (CSF) after epidural morphine injection in mongrel dogs, and to provide a reliable experimental basis for postoperative epidural injection of small doses of morphine.Methods Five healthy mongrel dogs were anesthetized with 0.1 mg / kg Morpolysin and the plasma and CSF concentrations were measured by HPLC at different time points, and the pharmacokinetic parameters were calculated by 3P87 software.Results: (P <0.01).
