本文采用免疫组织化学方法,在大鼠局部脑缺血灶,观察了细胞骨架神经丝蛋白200的变化。结果表明:缺血10分钟神经丝蛋白变化不明显;缺血30分钟,缺血灶内的染色变浅甚至部分染色缺失,并随缺血时间的延长,染色改变的程度愈重。在缺血早期细胞骨架神经丝蛋白200的改变,可能是脑缺血后神经元不可逆损伤的一个重要病理变化过程。此方法对脑缺血早期诊断优于常规的HE法,为脑保护药物的筛选提供了新的客观的形态学指标。
In this paper, we used immunohistochemistry to observe the changes of the cytoskeleton neurofilament protein 200 in the rat focal cerebral ischemia. The results showed that there was no obvious change of neurofilament proteins at 10 minutes after ischemia. The staining of ischemic lesions became lighter and even partially stained at 30 minutes after ischemia, and the staining changed more with the time of ischemia. Changes in the skeletal neurofilament protein 200 during the early phase of ischemia may be an important pathological process of neuronal irreversible damage after cerebral ischemia. This method is better than the conventional HE method for the early diagnosis of cerebral ischemia, which provides a new objective morphological index for the screening of brain protective drugs.
