目的探讨凋亡抑制基因survivin在喉鳞癌中的表达及其与Bcl-2、Bax基因表达的相关性。方法应用免疫组织化学链酶卵白素-生物素复合体(strept avidin-biotin complex,SABC)方法检测45例喉鳞癌及10例正常喉黏膜组织中survivin,Bcl-2及Bax基因的表达。结果喉鳞癌组织中,Survivin的表达率为80%,显著高于正常喉黏膜组织的表达率20%(P均<0.05)。Survivin蛋白表达与喉鳞癌的TNM分期及淋巴结转移有关(P均<0.05)。Survivin表达与Bcl-2蛋白表达呈正相关(P<0.05),而与Bax蛋白表达呈负相关(P<0.05)。结论Survivin在喉鳞癌的发生发展中起重要作用,可能成为喉鳞癌基因治疗的新靶点;survivin基因可能与凋亡相关基因Bcl-2、Bax在喉鳞癌的发展中分别起协同和拮抗作用。
Objective To investigate the expression of survivin gene in laryngeal squamous cell carcinoma and its relationship with Bcl-2 and Bax gene expression. Methods The expressions of survivin, Bcl-2 and Bax in 45 cases of laryngeal squamous cell carcinoma and 10 cases of normal laryngeal mucosa were detected by immunohistochemical streptavidin-biotin complex (SABC). Results The expression of Survivin in laryngeal squamous cell carcinoma was 80%, which was significantly higher than that in normal laryngeal mucosa (P <0.05). Survivin protein expression was correlated with TNM stage and lymph node metastasis of laryngeal squamous cell carcinoma (all P <0.05). Survivin expression was positively correlated with Bcl-2 protein expression (P <0.05), but negatively with Bax protein expression (P <0.05). Conclusions Survivin plays an important role in the development of laryngeal squamous cell carcinoma and may be a new target for gene therapy of laryngeal squamous cell carcinoma. Survivin may play a synergistic role in the development of laryngeal squamous cell carcinoma with the expression of survivin gene Bcl-2 and Bax, respectively Antagonism.
