大麦黄矮病毒(Barlearyellowdwarfvirus,BYDV)是小麦上发生的最重要的病毒,GPV株系是我国小麦黄矮病发生的主流株系。本研究将GPV株系3′端缺失的复制酶基因序列构建成用于转化小麦的植物表达载体,通过花粉管途径将目的基因转入小麦之中。T0及T1代的分子检测结果表明,目的基因已经整合到了小麦的染色体之中。获得的2个转基因系的T1代苗在温室中的初步抗病检测显示,转基因后代对GPV株系具有一定抗性。
Barley yellow dwarf virus (BYDV) is the most important virus in wheat and GPV strain is the mainstream of wheat yellow dwarf virus in China. In the present study, the deduced 3’-end replicase gene of GPV strain was constructed into a plant expression vector for transformation of wheat, and the target gene was transferred into wheat through the pollen tube pathway. T0 and T1 generation of molecular test results show that the target gene has been integrated into the chromosomes of wheat. The obtained preliminary disease resistance test of T1 generation of two transgenic lines in the greenhouse showed that the transgenic progeny had some resistance to the GPV strain.
