目的:研究植入用顺铂兔局部给药后生物样品中药物浓度的测定方法,并分别测定游离铂及总铂的浓度。方法:改进石墨炉原子吸收法的升温程序和上样方法;增加生物样品的超滤前处理方法,以去除顺铂的蛋白结合物便于测定游离铂含量;并按《中华人民共和国药典》的规定对测定方法进行验证和评估。结果:本测定方法的回收率为80%~110%,日内精密度(RSD)在5%以下,均符合《中华人民共和国药典》2010年版规定。组织中游离铂浓度约为总铂浓度的10%~30%,静脉血中游离铂浓度是总铂浓度的5%以下。结论:当前建立的石墨炉原子吸收方法能较好地测定出血液和组织样本中的游离铂和总铂浓度,且该方法无爆沸现象、重现性好、灵敏度高、数据准确。
OBJECTIVE: To study the determination of drug concentration in biological samples after local administration of cisplatin rabbit and determine the concentrations of free platinum and total platinum. Methods: To improve the temperature-programmed procedure and loading method of GFAAS; to increase the pretreatment method of biological samples by ultrafiltration to remove free platinum by removing the protein conjugate of cisplatin; and to determine the content of free platinum according to “The People’s Republic of China Pharmacopoeia” The method of measurement is validated and evaluated. Results: The recovery rate of this method was 80% ~ 110%, intraday precision (RSD) was below 5%, all in line with the 2010 edition of “Pharmacopoeia of the People’s Republic of China”. The free platinum concentration in the tissue is about 10% to 30% of the total platinum concentration, and the free platinum concentration in the venous blood is less than 5% of the total platinum concentration. CONCLUSION: The current established graphite furnace atomic absorption spectrometry is able to determine the free platinum and total platinum concentration in blood and tissue sample well. The method has no bump phenomenon, good reproducibility, high sensitivity and accurate data.
