目的 探讨面神经损伤及修复条件下 ,面神经核运动神经元STAT3mRNA和蛋白的变化规律。方法 采用原位杂交和免疫组化染色法 ,观察成年大鼠面神经切断和即刻端端吻合后面神经核STAT3mRNA和蛋白的时程变化。结果 面神经切断后损伤侧面神经核STAT3mRNA信号的强度增加 ,在术后第 2 1d和 35d ,面神经单纯切断组和面神经切断后端端吻合组间STAT3mRNA信号的强度相差显著(P <0 .0 5 )。结论 面神经损伤后 ,面神经核运动神经元内由STAT 3介导的细胞因子的信号转导增加。在面神经无再生状态下 ,STAT 3表达上调呈持续性 ;而在面神经再生状态下 ,随着面神经的成功再生 ,STAT 3可逐渐恢复至正常水平
Objective To investigate the changes of STAT3 mRNA and protein in facial motor neurons after facial nerve injury and repair. Methods The in situ hybridization and immunohistochemical staining were used to observe the temporal changes of STAT3 mRNA and protein in the facial nucleus following facial nerve transection and immediate end-to-end anastomosis in adult rats. Results The signal intensity of STAT3mRNA increased in facial nerve fibers after facial nerve transection. The difference of STAT3mRNA signal intensity between the facial nerve group and facial nerve group after 21d and 35d was significant (P <0.05) . Conclusion After facial nerve injury, signal transduction of STAT3-mediated cytokines is increased in motor neurons of facial nerve. In the condition of no facial nerve regeneration, the STAT3 expression up-regulated; in the condition of facial nerve regeneration, with the successful regeneration of the facial nerve, the STAT3 gradually returned to the normal level
