目的:探索菱叶红景天组织培养与植株再生技术并优化体外繁殖体系。方法:采用正交试验设计分别研究外植体种类、植物生长物质种类和浓度对菱叶红景天愈伤组织的诱导、丛生芽的产生以及生根诱导的影响,实验数据进行极差和方差分析。结果:影响菱叶红景天愈伤组织诱导的因素依次为外植体>2,4-D>6-BA,叶片产生愈伤组织最佳的培养基为MS附加2,4-D 1.5 mg.L-1和6-BA0.5 mg.L-1;影响菱叶红景天丛生芽诱导的因素依次为外植体>NAA>6-BA,茎段产生丛生芽最佳的培养基为MS附加6-BA1.5 mg.L-1和NAA1.0 mg.L-1;试管苗生根的最佳培养基为1/2MS附加IBA1.0 mg.L-1,生根率可达90%以上,生根试管苗移栽成活率达98%以上。结论:本试验初步建立了菱叶红景天组织培养与植株再生的基本体系。
Objective: To explore Rhodiola rosea tissue culture and plant regeneration technology and to optimize in vitro propagation system. Methods: Orthogonal design was used to study the effects of explant types, plant growth substances and concentrations on the induction of callus, shoot formation and rooting in Rhodiola sachalinensis. The experimental data were analyzed for variance and variance. Results: The factors affecting the induction of Rhodiola rosea were as follows: explants> 2,4-D> 6-BA, and the best medium for callus induction was MS supplemented with 2,4-D 1.5 mg.L -1 and 6-BA0.5 mg.L-1. The factors affecting the induction of cluster buds of Rhodiola rosea were: explants> NAA> 6-BA, and the best medium for stems to produce cluster buds was MS with 6- BA1.5 mg.L-1 and NAA1.0 mg.L-1. The optimal culture medium for rooting of test-tube plantlets was 1 / 2MS with IBA1.0 mg.L-1, the rooting rate could reach more than 90% Miao transplanting survival rate of 98% or more. Conclusion: This experiment initially established Rhodiola rosea tissue culture and plant regeneration of the basic system.
