目的:分析L-多巴脱羧酶(DDC)mRNA和蛋白在喉癌组织中的表达,以评估其在喉癌诊断和预后中的临床意义。方法:从53例喉癌患者的肿瘤切除手术中获取106份组织样品,并从这些组织样品中分别分离出总RNA。采用RT-PCR法对DDC mRNA的表达水平进行量化,同时采用Western Blot蛋白质印迹法对DDC蛋白的表达水平进行检测。结果:与正常喉黏膜组织相比,原发性喉癌(P=0.000)和复发性喉癌(P=0.033)样品中的DDC mRNA表达水平都出现了显著下调。且DDC mRNA表达水平与患者年龄和组织学分级无关,但与TNM分期之间存在显著的负相关性(P=0.021)。同时,我们在肿瘤和正常喉组织中都检测到了DDC蛋白的表达。结论:DDC的表达状态可能在喉癌的发展中发挥重要的作用,在未来的喉癌患者临床管理中具有重要的意义。
OBJECTIVE: To analyze the expression of L-dopa decarboxylase (DDC) mRNA and protein in laryngeal carcinoma to evaluate its clinical significance in the diagnosis and prognosis of laryngeal carcinoma. METHODS: Totally 106 tissue samples were obtained from resection of the tumor in 53 laryngeal cancer patients, and total RNA was isolated from each of these tissue samples. The expression of DDC mRNA was quantified by RT-PCR, and the expression of DDC protein was detected by Western Blot. Results: Compared with normal laryngeal mucosa, DDC mRNA expression levels in both primary laryngeal carcinoma (P = 0.000) and recurrent laryngeal carcinoma (P = 0.033) were significantly down-regulated. DDC mRNA expression level had no correlation with age and histological grade, but there was a significant negative correlation with TNM staging (P = 0.021). At the same time, we detected DDC protein expression in both tumor and normal laryngeal tissues. Conclusion: The expression of DDC may play an important role in the development of laryngeal cancer, which is of great significance in the future clinical management of patients with laryngeal cancer.
