本文报道用DTPA环酐(CDTPAA)连接113m于单克隆抗体(McAb)上的初步结果,并试验了不同比值的CDTPAA/McAb对标记率的影响。当此比值由1:20增至1:200时。标记率由61%升至68%。经与CDTPAA水解对照组比较,对照组的标记率仅为25%。本结果提示,CDTPAA与McAb的结合是以未水解的CDTPAA连接放射性金属元素于McAb上为基础的。作者还讨论了反应环境中的pH值、反应时间及蛋白质浓度对连接效率的影响。本文对放射免疫治疗有意义的金属元素的标记法做了有益探索。
This paper reports the preliminary results of the ligation of 113m with monoclonal antibody against DTPA (CDTPAA) and the effect of different ratios of CDTPAA / McAb on the labeling efficiency. When this ratio increased from 1:20 to 1: 200. Marking rate increased from 61% to 68%. Compared with the CDTPAA hydrolysis control group, the labeling rate of the control group was only 25%. The results suggest that the binding of CDTPAA to McAbs is based on McAb by linking unlabeled CDTPAA to radioactive metal elements. The authors also discuss the effect of pH, reaction time and protein concentration on the efficiency of the reaction in the reaction environment. This article makes a useful exploration of the labeling of metallic elements that are of interest in radioimmunotherapy.
