目的:观察内肥素对THP-1源性泡沫细胞形成中血凝素样氧化低密度脂蛋白受体-1(LOX-1)表达的影响,及其在动脉粥样硬化中的作用机制。方法:给予不同浓度的内肥素处理24h和同一浓度的内肥素处理THP-1源性巨噬细胞0~36h,运用油红O染色观察细胞内脂滴的变化,运用逆转录聚合酶链反应和Western-blot检测LOX-1的mRNA和蛋白的表达水平。结果:高浓度的内肥素处理负荷LDL的THP-1源性巨噬细胞24h后,细胞浆内的脂滴明显增多。在负荷LDL的THP-1源性巨噬细胞上,内肥素上调LOX-1mRNA和蛋白表达。结论:内肥素上调THP-1源性泡沫细胞中LOX-1的表达。
Objective: To investigate the effect of endogenous lipoxin on the expression of LOX-1 in THP-1-derived foam cells and its mechanism in atherosclerosis. Methods: The THP-1-derived macrophages were treated with different concentrations of endotoxin for 24 h and the same concentration of endotoxin for 0-36 h. The changes of intracellular lipid droplets were observed by oil red O staining. The expression of LOX-1 mRNA and protein was detected by Western blot and Western blot. RESULTS: High concentration of endogenous liposomes significantly increased lipid droplets in cytoplasm after treatment with THP-1-derived macrophages loaded with LDL for 24 h. On LDL-loaded THP-1-derived macrophages, endocytosis up-regulates LOX-1 mRNA and protein expression. CONCLUSION: Endostatin up-regulated LOX-1 expression in THP-1-derived foam cells.
