目的 观察去卵巢大鼠大脑皮质和海马突触体[Ca2+]i和膜流动性变化以及染料木素的治疗作用。 方法 雌性SD大鼠随机分为假手术组、去卵巢对照组、染料木素组、苯甲酸雌二醇组。分别皮下注射生理盐水 50μ1、生理盐水50μ1、染料木素250μg和苯甲酸雌二醇50μg,隔日1次。术后8周测定各组大鼠大脑额、顶叶皮质 和海马的突触体[Ca2+]i及膜流动性。 结果 去卵巢对照组大鼠大脑皮质及海马突触体[Ca2+]i分别为(243.31 ±31.21)nmol/L和(305.10±54.31)nmol/L,与假手术组、苯甲酸雌二醇组和染料木素组之间有显著性差异(P< 0.01);但在假手术组、苯甲酸雌二醇组和染料木素组之间无显著差异(P>0.05);去卵巢对照组海马突触体膜相对 黏滞度(η)为3.03±0.39,与假手术组和苯甲酸雌二醇组之间有显著性差异(P<0.05);与染料木素组相比有高度 显著性差异(P<0.01)。染料木素组、苯甲酸雌二醇组及假手术组之间无显著差异(P>0.05)。 结论 大鼠去卵 巢后突触体膜流动性降低及突触体内自由钙增加,染料木素能增加去卵巢大鼠突触体膜流动性及维持钙稳态。
Objective To observe the changes of [Ca2 +] i and membrane fluidity in the cortex and hippocampus of ovariectomized rats and the therapeutic effect of genistein. Methods Female Sprague-Dawley rats were randomly divided into sham operation group, ovariectomized group, genistein group and estradiol benzoate group. Subcutaneous injection of normal saline 50μ1, saline 50μ1, genistein 250μg and estradiol benzoate 50μg, every other day. At 8 weeks after operation, the synaptosomal [Ca2 +] i and membrane fluidity of the frontal cortex and parietal cortex and hippocampus of rats in each group were determined. Results The [Ca2 +] i of cerebral cortex and hippocampus in the ovariectomized group were (243.31 ± 31.21) nmol / L and (305.10 ± 54.31) nmol / L, respectively. Compared with sham operation group, estradiol benzoate group and There was a significant difference between genistein group and genistein group (P <0.01), but there was no significant difference between sham operation group, estradiol benzoate group and genistein group (P> 0.05) The relative viscosity (η) of the synovial membrane was 3.03 ± 0.39, which was significantly different from that of the sham operation group and the estradiol benzoate group (P <0.05). Conclusions After ovariectomy, the fluidity of synaptosomal membrane is decreased and free calcium is increased in synaptosome. Genistein can increase the fluidity of synaptosomal membrane and maintain the calcium homeostasis in ovariectomized rats.
