目的观察不同培养基对人脐带间充质干细胞(hUC-MSCs)生物学特性的影响,优化培养方法。方法采用贴壁法从正常足月胎儿脐带中分离出间充质干细胞,细胞贴壁后利用DMEM/F12,Mesen PRORSTM Medium这两种培养体系进行体外扩增,绘制生长曲线,流式细胞仪检测其表面标志。结果从人脐带中分离出的间充质干细胞为梭形,呈平行排列生长或漩涡状生长;细胞表达CD29、CD44,不表达CD34、CD45和HLA-DR。结论加入Mesen PRORSTM Medium培养的细胞增殖速度快,更适合于脐带间充质干细胞的体外扩增。
Objective To observe the effects of different culture medium on the biological characteristics of human umbilical cord mesenchymal stem cells (hUC-MSCs) and to optimize the culture method. METHODS: Mesenchymal stem cells (MSCs) were isolated from umbilical cord of normal full-term fetuses by adherence method. After adherent cells, the cells were expanded in vitro using DMEM / F12 and Mesen PRORSTM Medium, and the growth curves were drawn. Flow cytometry Its surface logo. Results The mesenchymal stem cells isolated from human umbilical cord were spindle-shaped and grew parallel or swirled. The cells expressed CD29 and CD44 but did not express CD34, CD45 and HLA-DR. Conclusion The cells cultured in Mesen PRORSTM Medium can proliferate rapidly and are more suitable for in vitro expansion of umbilical cord mesenchymal stem cells.
