目的:探讨环氧化酶-2基因(COX-2)的两个功能性单核苷酸多态(Single nucleotide polymorphisms,SNPs)与胃癌易感性的关系。方法:采用病例-对照研究设计,研究对象为254例经组织学确诊的胃癌病例和304例按年龄、性别进行匹配的无肿瘤史的社区对照人群。采用聚合酶链反应-限制性片段长度多态性方法(PCR-RFLP)测定COX-2基因-1195G/A多态,错配聚合酶链反应-限制性片段长度多态性方法(PIRA-PCR)测定COX-2基因8473T/C多态的基因型,比较不同基因型与胃癌风险的关系,并探讨不同环境因素在其中所起的影响。结果:COX-2-1195GG、GA和AA三种基因型以及8473TT、TC和CC三种基因型在病例组和对照组间的频率分布无统计学意义。Logistic回归分析也未发现COX-2 SNP-1195G/A以及SNP8473T/C与胃癌的风险度关联有统计学意义(SNP-1195 GA+AA:校正OR=1.09,95%CI=0.70-1.67;SNP8473 TC+CC:校正OR=1.18,95%CI=0.83-1.69)。结论:COX-2-1195G/A和8473T/C多态性可能与胃癌易感性无关,尚需进一步大样本量的研究予以证实。
Objective: To investigate the relationship between two single nucleotide polymorphisms (SNPs) of cyclooxygenase-2 (COX-2) gene and susceptibility to gastric cancer. METHODS: A case-control study was designed. The study population consisted of 254 patients with histologically diagnosed gastric cancer and 304 age-matched and sex-matched community-free controls. Polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) was used to determine COX-2 gene-1195G / A polymorphism and mismatched polymerase chain reaction-restriction fragment length polymorphism ) Was used to determine the genotype of the 8473T / C polymorphism in COX-2 gene. The relationship between different genotypes and the risk of gastric cancer was compared and the influence of different environmental factors was explored. Results: The frequency distribution of COX-2-1195GG, GA and AA genotypes and 8473TT, TC and CC genotypes between the case group and the control group were not statistically significant. Logistic regression analysis also showed no significant correlation between COX-2 SNP-1195G / A and SNP8473T / C and risk of gastric cancer (SNP-1195GA + AA: corrected OR = 1.09,95% CI = 0.70-1.67; SNP8473 TC + CC: corrected OR = 1.18, 95% CI = 0.83-1.69). Conclusion: The polymorphisms of COX-2-1195G / A and 8473T / C may not be related to the susceptibility to gastric cancer. However, further studies with large sample size are needed to confirm this.
