目的:分离大鼠皮层乙酰胆碱酯酶G4和G1分子亚型以阐明乙酰胆碱酯酶抑制剂药物的作用。方法:使用分子排阻层析和超速离心方法分离和确证乙酰胆碱酯酶的G4和G1分子亚型.结果:经过分子排阻层析得到两个乙酰胆碱酯酶的活性峰,与标准蛋白质的洗脱峰相比,推测其分子量为M_r 239000和M_r 68000.通过超速离心法确证其沉降系数为10S和4S,分别为乙酰胆碱酯酸G4和G1分子亚型.结论:乙酰胆碱酯酶的G4和G1分子亚型可以使用分子排阻层析的方法分离得到.
OBJECTIVE: To separate the acetylcholinesterase G4 and G1 subtypes in rat cortex to elucidate the effects of acetylcholinesterase inhibitors. Methods: The G4 and G1 subtypes of acetylcholinesterase were isolated and confirmed by size exclusion chromatography and ultracentrifugation.Results: Two acetylcholinesterase activity peaks were obtained by molecular exclusion chromatography, Peak, the molecular weights of which were estimated to be M_r 239000 and M_r 68000. The sedimentation coefficients were confirmed to be 10S and 4S by ultracentrifugation, respectively, which were acetylcholinesterase G4 and G1 subtypes.Conclusion: The acetylcholinesterase G4 and G1 molecular sub The type can be isolated using the method of size exclusion chromatography.
