背景:脐血单个核细胞可作为种子细胞源以移植修复不同原因所致脑功能损害,但能否表达神经元另一重要特有分子τ蛋白?目的:探讨脐血单个核细胞在培养前后及细胞因子诱导作用下,τ蛋白mRNA的表达。设计:随机对照研究。地点和对象:实验地点:华中科技大学同济医学院。采集健康新生儿脐血8份,每份80mL。干预:在细胞培养过程中,使用细胞因子:表皮生长因子(epidermalgrowthfactor,EGF)和碱性成纤维细胞生长因子(basicfibroblastgrowthfactor,bFGF)。主要观察指标:①细胞形态。②τ蛋白mRNA在脐血单个核细胞中的表达。结果:培养前,脐血单个核细胞胞体小呈圆形,培养后EGF+bFGF组细胞胞体较大,有粗长突起形成,未加细胞因子所培养细胞胞体较小,突起较短、较细,形似星形细胞。反转录聚合酶链式反应(reverse-transcriptpolymerasechainreaction,RT-PCR)检测未培养的脐血单个核细胞τ蛋白mRNA呈阴性而MAP2mRNA表达阳性,培养后二者均表达阳性,在EGF+bFGF作用下,阳性表达更明显。结论:脐血单个核细胞表达MAP2mRNA,经培养后同时表达τ蛋白mRNA,细胞因子能增强这种作用。
BACKGROUND: Umbilical cord blood mononuclear cells can be used as a seed cell source for transplantation to repair brain damage caused by different reasons, but whether it can express another important characteristic molecule of tau protein in neurons.Objective: To investigate whether umbilical cord blood mononuclear cells Factor-induced expression of tau protein mRNA. Design: Randomized controlled study. Venue and Subject: Experiment Location: Tongji Medical College, Huazhong University of Science and Technology. Eight healthy neonatal cord blood samples were collected, each 80mL. Interventions: Cytokines such as epidermal growth factor (EGF) and basic fibroblast growth factor (bFGF) are used during cell culture. MAIN OUTCOME MEASURES: ① cell morphology. ② Tau protein mRNA expression in cord blood mononuclear cells. Results: Before culture, the cord blood mononuclear cells were small and round in shape. After culturing, the cells of EGF + bFGF group had large cell body with long protuberances. Cells cultured without cytokines were smaller, the protuberances were shorter and thinner , Shaped like a star cell. Reverse transcript polymerase chain reaction (reverse-transcript polymerase chain reaction, reverse transcriptase polymerase chain reaction (RT-PCR) detection of umbilical cord blood mononuclear cells in tau protein mRNA was negative and MAP2 mRNA expression was positive after incubation were expressed in EGF + bFGF , The positive expression is more obvious. CONCLUSION: MAP2 mRNA is expressed in umbilical cord blood mononuclear cells and expressed simultaneously by tau mRNA after culture. Cytokines can enhance this effect.
