目的:研究免疫球蛋白对新生大鼠缺血缺氧性脑损伤(HIBD)时炎症因子抵抗素、白细胞介素-1β(IL-1β)和NF-κB p65表达的影响。方法:建立HIBD模型;将72只新生7日龄SD仔鼠随机分为3组:假手术组(n=24),HIBD组(n=24,腹腔内注射生理盐水,0.02 ml·g-1·次-1,2次/d,共2天),治疗组(n=24,腹腔内注射免疫球蛋白,2.5 mg·g-1·次-1,2次/d,共2天)。每个组按照12 h、24 h、72 h时间点分为3个亚组,每亚组8只;以免疫组化法检测病变侧脑组织中抵抗素、IL-1β和NF-κB p65的水平。结果:抵抗素、IL-1β及NF-κB p65在假手术组均只有少量表达,各时间点之间比较差异无统计学意义(P>0.05);抵抗素、IL-1β及NF-κB p65在HIBD组12 h表达开始增高,24 h达到高峰,随时间进展,其表达量逐渐减少;与假手术组各时间点相比,HIBD组和治疗组抵抗素、IL-1β及NF-κB p65表达均增高,差异有统计学意义(P<0.05);与HIBD组相比,治疗组12 h、24 h、72 h抵抗素、IL-1β及NF-κB p65表达量减少,差异有统计学意义(P<0.05)。结论:NF-κB p65、抵抗素及IL-1β在HIBD大鼠中表达上调,免疫球蛋白能下调NF-κB p65、抵抗素及IL-1β的表达。
Objective: To investigate the effect of immunoglobulin on the expression of resistin, interleukin-1β (IL-1β) and NF-κB p65 in neonatal rats with hypoxic-ischemic brain damage (HIBD). Methods: Seventy-two neonatal SD rats of 7 days old were randomly divided into 3 groups: sham operation group (n = 24), HIBD group (n = 24, intraperitoneal injection of saline, 0.02 ml · g -1 (N = 24, intraperitoneal injection of immunoglobulin, 2.5 mg · g -1 · -1,2 times / d for 2 days). Each group was divided into 3 subgroups according to the time points of 12 h, 24 h and 72 h, and each subgroup was divided into 8 subgroups. Compared with HIBD group, the expression of resistin, IL-1β and NF-κB p65 decreased at 12 h, 24 h, 72 h in the treatment group, the differences were statistically significant Significance (P <0.05). CONCLUSION: The expressions of NF-κB p65, resistin and IL-1β are up-regulated in HIBD rats. Immunoglobulin can down-regulate the expressions of NF-κB p65, resistin and IL-1β.
