目的 研究胃泌素 (Gastrin ,GAS)、生长抑素 (Somatostatin ,SS)在大肠癌 (Colorectalcarcinoma)组织的表达变化以及与大肠癌细胞增殖指数、凋亡指数的关系 ,探讨大肠癌胃泌素、生长抑素的表达与大肠癌细胞增殖 (Cellproliferation)、细胞凋亡 (Apoptosis)之间的相关性 ,为大肠癌的内分泌治疗提供依据。 方法 收集大肠癌 (结、直肠癌 )标本 60例 ,取癌组织用中性福尔马林固定 ,石蜡包埋 ,作 4μm厚连续切片 ,用免疫组织化学方法中的链霉抗生物素蛋白 生物素 过氧化物酶复合体法 (SABC)和分子生物学细胞原位凋亡检测技术中的以DNA末端转移酶介导的缺口末端标记法 (TUNEL) ,检测GAS、SS、增殖细胞核抗原 (PCNA)、Ki 67和凋亡细胞的表达情况。结果 ①在大肠癌组织GAS高表达组PI、KI的表达高于GAS低表达组 ,而AI的表达则相反。②AI在大肠癌组织SS高表达组的表达高于SS低表达组 ,而PI、KI的表达则相反。③大肠癌组织中GAS与SS表达的二项指数积分比值与细胞增殖呈明显正相关 ,与细胞凋亡呈显著负相关。结论 ①在大肠癌组织中 ,GAS表达的水平越高 ,细胞增殖活性愈高 ,凋亡指数却越低 ;而SS表达的水平愈高 ,凋亡指数越高 ,细胞增殖活性却愈低。大肠癌组织GAS/SS的比值变化与细胞增殖呈正相关 ,和细胞凋亡呈?
Objective To investigate the expression of gastrin (GAS) and somatostatin (SS) in colorectal carcinoma tissue and the relationship with the proliferation index and apoptosis index of colorectal cancer cells, and to explore the correlation between the expression of gastrin, Somatostatin expression and colorectal cancer cell proliferation (Cellproliferation), apoptosis (Apoptosis) correlation between the endocrine therapy for colorectal cancer provide the basis. Methods Sixty specimens of colorectal cancer (colorectal cancer) were collected. The cancerous tissues were fixed with formalin, embedded in paraffin and cut into 4μm thick sections. The streptavidin biosynthesis Peroxidase complex assay (SABC) and molecular biological detection of in situ cell death by DNA terminal transferase mediated nick end labeling (TUNEL), detection of GAS, SS, proliferating cell nuclear antigen (PCNA ), Ki 67 and apoptotic cells. Results ① The expression of PI and KI in GAS overexpression group was higher than that in GAS overexpression group, while the expression of AI was the opposite in colorectal cancer tissues. ② The expression of AI in SS high expression group was higher than that in SS low expression group, while the expression of PI and KI was the opposite. ③ The ratio of binomial index of GAS and SS expression in colorectal carcinoma was positively correlated with cell proliferation, and significantly negatively correlated with apoptosis. Conclusions ① The expression of GAS is higher in colorectal cancer tissues, the higher the cell proliferation activity is, the lower the apoptosis index is. However, the higher the level of SS expression, the higher the apoptosis index and the lower the cell proliferation activity.
