目的优选祛毒清热颗粒中挥发油的提取方法,确定其β-环糊精的包合工艺。方法按《中国药典》2010年版挥发油提取方法提取挥发油,以提取率为指标,对影响挥发油提取的因素(加水量、药材粉碎程度、挥发油提取时间)进行考察;以包合物的收率和油转化率的综合评分为指标,应用L9(34)正交试验进行包合条件的优化。结果挥发油最佳提取工艺为药材粉碎过20目筛,加7倍量水,提取4 h。β-环糊精的最佳包合工艺为β-环糊精与油的质量比为8︰1,包合温度40℃,包合时间为2 h,无水乙醇与挥发油的体积比为1︰3。结论该优选提取和包合工艺合理可行,可作为祛毒清热颗粒中挥发油β-环糊精包合物的制备。
Objective To optimize the method of extracting volatile oil from Qudu Qingre granules and determine the inclusion process of β-cyclodextrin. Methods According to the “Chinese Pharmacopoeia” 2010 edition of volatile oil extraction method to extract volatile oil, extraction rate as an indicator, the factors that affect the extraction of volatile oil (water, medicine grinding degree, volatile oil extraction time) were investigated; inclusion compound yield and oil Conversion rate of the composite score as an indicator, the application of L9 (34) orthogonal test inclusion conditions optimization. Results The best extraction process of volatile oil was pulverized 20 mesh sieve, add 7 times the amount of water, extracted 4 h. β-cyclodextrin inclusion process is β-cyclodextrin and oil mass ratio of 8: 1, Inclusion temperature 40 ℃, Inclusion time is 2 h, the volume ratio of absolute ethanol and volatile oil is 1 ︰3. Conclusion The optimal extraction and inclusion process is reasonable and feasible, which can be used as the preparation of β-cyclodextrin inclusion complex of volatile oil in Qudu Qingre granules.
