本实验分别以ESR自旋标记法、TBA比色法测定大鼠离体心脏缺血,再灌注时线粒体膜流动性及脂质过氧化物含量。发现心肌线粒体膜深层的流动性在缺血40分钟后有氧再灌注20分钟时(S=0.112±0.006、τ_e=7.13±0.09×10~(-10)sec)明显低于富氧灌注组(S=0.103±0.007、τ_e=6.86±0.20×10~(-10)sec),P<0.05;而脂质过氧化物含量再灌注组(3.02±0.68nmol/mgpr.)则极明显高于富氧灌注组(1.94±0.35nmol/mgpr.),P<0.01。预先给予丹参素,对以上变化有明显的改善作用,上述两项指标与富氧灌注组相比,无明显差别,P>0.05。另外,采用ESR自旋捕集技术发现丹参素对外源性O_2~-·有清除作用。因而推测,丹参素可能作为一种O_2~-·的清除剂保护心肌线粒体免受氧自由基引发的脂质过氧化损害。
In this study, the mitochondrial membrane fluidity and lipid peroxidation of rat isolated heart were detected by ESR spin-labeling and TBA colorimetry, respectively. It was found that the deep fluidity of myocardial mitochondrial membrane was significantly lower than that of oxygen-enriched perfusion group (P <0.05) after 20 minutes of reperfusion of oxygen 40 minutes after ischemia (S = 0.112 ± 0.006, τ_e = 7.13 ± 0.09 × 10 ~ (P <0.05). However, the level of lipid peroxidation in the reperfusion group (3.02 ± 0.68nmol / mgpr.)
