目的探讨全反式维甲酸对骨髓基质细胞(bone marrow stromal cells,BMSC)黏附功能及JWA基因表达的影响。方法全反式维甲酸(all trans retinoic acid,atRA)作用BMSC后,流式细胞仪检测BMSC细胞间黏附分子-1(intercellular adhe-sion molecule-1,ICAM-1)和血管黏附分子-1(vascular adhesion molecule-1,VCAM-1)的表达,检测共培养条件下BMSC细胞对Jurkat细胞的黏附率及Jurkat细胞生长曲线变化,RT-PCR方法检测BMSC JWA mRNA表达变化。结果atRA作用后,BMSC表面ICAM-1、VCAM-1蛋白表达增加,BMSC对Jurkat细胞的黏附能力增强,并可促进Jurkat细胞增殖,BMSC内JWA-mRNA表达增强。结论atRA可增强骨髓基质细胞黏附功能,并上调JWA基因表达。
Objective To investigate the effect of all-trans retinoic acid on the adhesion of bone marrow stromal cells (BMSC) and the expression of JWA gene. Methods After all-trans retinoic acid (atRA) treated with BMSC, the expressions of intercellular ad-sion molecule-1 (ICAM-1) and vascular cell adhesion molecule-1 Vascular endothelial growth factor (VCAM-1) and vascular adhesion molecule-1 (VCAM-1) were detected by flow cytometry. The adhesion rate of Jurkat cells and the growth curve of Jurkat cells in BMSC cells were detected under co-culture conditions. Results After treated with atRA, the expression of ICAM-1 and VCAM-1 on BMSC increased, and the adhesive ability of BMSC to Jurkat cells increased. The proliferation of Jurkat cells was enhanced and the expression of JWA-mRNA in BMSC was enhanced. Conclusions atRA can enhance the adhesion of bone marrow stromal cells and up-regulate the expression of JWA gene.
