为探讨1例急性粒单核细胞白血病原始细胞表达血小板特异性抗原的机制,分离外周血或骨髓单个核细胞,采用常规染色和普通细胞化学染色观察其形态学特性,采用流式细胞仪双荧光分析免疫表型,单荧光胞浆蛋白标记分析周期蛋白表达和免疫印迹进一步证实周期蛋白D_3的表达情况,采用周期蛋白和DNA双标记分析周期蛋白D_3表达与细胞周期的关系。结果表明,在白血病原始细胞中CD_(13)~++HLA-DR~+细胞为94.69%,CD_(13)~++CD_(34)~+细胞为96.86%,CD_(41a)~++CD_(34)~+细胞为41.6%,CD_(13)~++CD_(41a)~+细胞为40.0%;周期蛋白D_3表达明显升高,大部分细胞表达周期蛋白D_3,周期蛋白D_3阳性细胞在细胞周期各时段所占的比例分别为:G_1期52.1%,S期9.9%,G_2+M期38.0%。结论:周期蛋白D_3的异常表达导致此白血病细胞表达血小板特异性抗原,可能在白血病的发生中起重要的作用。
To investigate the mechanism of expression of platelet-specific antigen in a promyelocytic leukemia blast cell line, peripheral blood or bone marrow mononuclear cells were isolated and their morphological characteristics were observed by conventional staining and ordinary cytochemical staining. Flow cytometry dual fluorescence was used. The immunophenotype was analyzed, and the expression of cyclin D3 was further confirmed by the expression of cyclin protein and Western blotting. The relationship between the expression of cyclin D3 and the cell cycle was analyzed by cyclin and DNA double labeling. The results showed that CD_(13)~++HLA-DR~+ cells were 94.69% in CD3+CD_(34)~+cells, and CD_(41a)~++ The number of CD_(34)~+ cells was 41.6%, and that of CD_(13)~++CD_(41a)~+ cells was 40.0%. The expression of cyclin D_3 was significantly increased. Most cells expressed cyclin D_3 and cyclin D_3 positive cells. The proportions of the cell cycle in each period were: 52.1% in G 1 phase, 9.9% in S phase, and 38.0% in G 2 +M phase. CONCLUSION: The abnormal expression of cyclin D3 leads to the expression of platelet-specific antigen in leukemia cells, which may play an important role in the development of leukemia.
