为探讨兔下颌骨髁状突软骨细胞的体外培养方法及其生物学特性 ,用胰酶及胶原酶联合消化法获取软骨细胞 ,并在DMEM培养基中进行原代及传代培养 ;通过形态学观察及免疫组织化学染色对软骨细胞的贴壁率、生长曲线及表型等细胞生物学特性进行了研究 .结果 :所获软骨细胞主要呈多角形 ,第 3代软骨细胞 12h贴壁率达 95% ,软骨细胞倍增时间约 55 1h ,6代以内软骨细胞的II型胶原蛋白稳定表达 .结果表明 :本方法所获软骨细胞具有稳定、良好的生长特性 .
To explore the culture method and biological characteristics of rabbit mandibular condyle chondrocytes in vitro, chondrocytes were obtained by digestion with trypsin and collagenase and cultured in DMEM medium for primary culture and subculture. Morphological observation And chondrocytes adherent rate, growth curve and phenotype were studied by immunohistochemical staining.Results: The chondrocytes were mainly polygons, the third generation of chondrocytes adherence rate of 12h 95% , The chondrocyte doubling time was about 55 1 h, and the expression of type II collagen in chondrocytes was stable within 6 generations.The results showed that the chondrocytes obtained by this method have stable and good growth characteristics.
