研究黄连素对恶性胶质瘤C6细胞增殖、凋亡的影响.用MTT法分别检测不同浓度的黄连素对恶性胶质瘤C6细胞体外抑制率.通过不同时间、不同浓度的黄连素对胶质瘤C6细胞的作用,于倒置相差显微镜下观察C6细胞的形态变化,以及Western Blot检测PARP的特异性切割.MTT实验结果表明黄连素对恶性胶质瘤C6细胞生长的抑制率随药物浓度和作用时间的增加而增加(P<0.05).除了在48 h,药物浓度63.7μmol/L,191μmol/L之间的抑制率无差异(P>0.05),其余不同浓度之间具有统计学意义.黄连素作用下C6细胞变圆、胞质回缩、贴壁细胞悬浮脱落,细胞发生凋亡,进一步Western Blot检测PARP发生特异性切割.黄连素对恶性胶质瘤C6细胞体外增殖有抑制作用,同时诱导细胞凋亡.本研究证明黄连素有一定的抑癌作用,可望成为一种新型的抗胶质瘤药.
To study the effect of berberine on the proliferation and apoptosis of C6 glioma cells.MTT assay was used to detect the inhibitory rate of different concentrations of berberine on C6 glioma cells in vitro.Under different concentrations of berberine, C6 cells were observed under inverted phase contrast microscope to observe the morphological changes of C6 cells and the specific cleavage of PARP detected by Western Blot.The MTT results showed that the inhibitory rate of berberine on the growth of malignant glioma C6 cells with the drug concentration and effect (P <0.05) .At 48 h, no significant difference was found between the inhibition rates of drug concentration of 63.7μmol / L and 191μmol / L (P> 0.05) C6 cells became round, cytoplasm was retracted, cells were suspended and detached, apoptosis of cells was detected by Western Blot, and PARP was detected by Western Blot.Cytisine inhibited the proliferation of C6 glioma cells in vitro, Induced apoptosis.It is proved that berberine has a certain anti-cancer effect and is expected to become a new type of anti-glioma drug.
