目的:利用我室建立的视网膜母细胞瘤细胞株SO-Rb_(50)和SO-Rb_(70)探索MTT法用于悬浮生长的RB细胞的有关因素,及骆驼蓬总碱对这两个瘤细胞株体外抑制作用。方法:分别测试瘤细胞数与OD值(光密度值)的关系,MTT作用时间与OD值的关系,及结晶物经DMSO(二甲基亚砜)溶解后的稳定时间,并用MTT法检测了骆驼蓬总碱对这两个瘤细胞株体外抑制效果。结果:这两个瘤细胞株活细胞数量与OD值成正比,MTT作用时间在12小时内OD值渐增加,结晶物经DMSO溶解后的稳定时间为11小时。骆驼蓬总碱作用于SO-Rb_(50)瘤细胞48和72小时的IC50值(μg/ml)分别为10.66和4.82;作用于SO-Rb_(70)瘤细胞48和72小时的IC50值(μg/ml)分别为6.37和4.2。结论:MTT法可用于悬浮生长的Rb细胞,骆驼蓬总碱对SO-Rb_(50)和SO-Rb_(70)瘤细胞有明显的抑制作用。眼科学报 1996;12:83—87。
OBJECTIVE: To explore the relevant factors of RB cells used for suspension growth by MTT method and SO2-Rb_ (50) and SO-Rb_ (70) of retinoblastoma cell lines established in our laboratory, Cell lines in vitro inhibition. Methods: The relationship between the number of tumor cells and the OD value (optical density value), the relationship between the MTT action time and the OD value and the stability time of the crystal after the DMSO (dimethylsulfoxide) dissolution were tested. The MTT assay Peganum harmala total alkaloids inhibit the two tumor cell lines in vitro. RESULTS: The number of viable cells in these two tumor cell lines was proportional to the OD value. The OD value of MTT increased gradually within 12 hours, and the stability time of the crystals dissolved in DMSO was 11 hours. IC50 values (μg / ml) of harmala alkaloids on SO-Rb_ (50) tumor cells at 48 and 72 hours were 10.66 and 4.82, respectively; IC50 values at 48 and 72 hours μg / ml) were 6.37 and 4.2, respectively. CONCLUSION: MTT can be used for the growth of Rb cells in suspension, and total alkaloids of Peganum harma have obvious inhibitory effects on SO-Rb 50 and SO-Rb 70 tumor cells.
