目的:探讨p-mTOR、p-4EBP1在横纹肌肉瘤组织中的表达及临床意义,并探讨其相关性。方法:采用免疫组化(SABC法)检测70例横纹肌肉瘤组织p-mTOR、P-4EBPl的表达,分析其与横纹肌肉瘤患者临床病理特征及预后的关系。结果横纹肌肉瘤组织中p-mTOR、p-4EBP1蛋白阳性表达率分别为75.7%和65.7%,均呈正相关(P<0.05)。p-mTOR蛋白表达与其病理亚型相关性有统计学意义(P<0.05),p-mTOR蛋白阴性组患者3年总生存率、无疾病进展生存率均优于阳性组患者(P=0.001,P=0.014)。P-4EBPl蛋白表达与患者的肿瘤大小的相关性有统计学意义(P<0.05),p-4EBP1蛋白阴性组患者3年总生存率、无疾病进展生存率均优于阳性组患者(P=0.017,P=0.052)。单因素分析显示,p-mTOR、p-4EBP1、分期、病理类型、肿瘤大小是不良预后因素(P<0.05),而淋巴结转移情况、性别、年龄与预后无关(P>0.05)。结论:横纹肌肉瘤组织中存在p-mTOR、p-4EBPl蛋白的高表达与肿瘤临床特点有关,p-mTOR、p-4EBP1的表达存在相关性,p-mnTOR和p-4EBP1表达是横纹肌肉瘤的不良预后因素。
Objective: To investigate the expression of p-mTOR and p-4EBP1 in rhabdomyosarcoma and their clinical significance, and to explore their correlation. Methods: The expressions of p-mTOR and P-4EBP1 in 70 cases of rhabdomyosarcoma were detected by immunohistochemistry (SABC method), and their relationship with the clinicopathological features and prognosis of rhabdomyosarcoma were analyzed. Results The positive rates of p-mTOR and p-4EBP1 protein in rhabdomyosarcoma tissues were 75.7% and 65.7%, respectively (P <0.05). The correlation between the expression of p-mTOR protein and its pathological subtype was statistically significant (P <0.05). The 3-year overall survival and disease-free survival rate of patients with p-mTOR negative were significantly higher than those of patients with positive p-mTOR P = 0.014). The correlation between the expression of P-4EBP1 protein and tumor size was statistically significant (P <0.05). The 3-year overall survival and disease-free survival rate in patients with p-4EBP1 negative were significantly higher than those in patients with positive (P = 0.017, P = 0.052). Univariate analysis showed that p-mTOR, p-4EBP1, stage, pathological type and tumor size were negative prognostic factors (P <0.05), while lymph node metastasis, gender and age had no relation with prognosis (P> 0.05). CONCLUSIONS: The high expressions of p-mTOR and p-4EBP1 in rhabdomyosarcoma tissues are related to the clinical features of the tumor. The expressions of p-mTOR and p-4EBP1 are correlated. The expressions of p-mnTOR and p-4EBP1 are abnormal in rhabdomyosarcoma Prognostic factors.
