目的:分离纯化及鉴定一种新的乳腺癌相关抗原。方法:将乳腺癌组织粗提取物进行DEAE-纤维素阶段洗脱,经ELISA法检测其特异性蛋白峰,通过SDS-PAGE电泳和ELISA方法进一步纯化及鉴定该抗原,并用酶法、热稳定性检测其理化特性。结果:洗脱的3个蛋白峰中,峰1与乳腺癌患者血清中抗体反应,不与健康妇女、良性乳腺疾病和其它肿瘤患者的血清反应。乳腺癌相关抗原存在于峰1中,分子量为85kDa,为糖蛋白。结论:该抗原在乳腺癌的诊断及判断预后方面具有一定的价值。
Objective: To isolate, purify and identify a new breast cancer related antigen. Methods: The crude extract of breast cancer tissue was subjected to DEAE-cellulose phase elution. The specific protein peak was detected by ELISA. The antigen was further purified and identified by SDS-PAGE electrophoresis and ELISA. The enzymatic activity, thermostability Test its physical and chemical properties. Results: In the three protein peaks eluted, peak 1 reacted with the antibody in serum of breast cancer patients and did not react with sera of healthy women, benign breast diseases and other cancer patients. The breast cancer associated antigen is present in peak 1 and has a molecular weight of 85 kDa, which is a glycoprotein. Conclusion: This antigen has some value in diagnosis and prognosis of breast cancer.
