目的:观察大蓟醇提物对两肾一夹(2-kidney 1-clip,2K1C)高血压大鼠的影响及其机制。方法:采用2K1C复制肾性高血压大鼠模型,用无创血压测量系统测定给予大蓟醇提物前后大鼠尾动脉压的变化,放射免疫学方法检测血浆中血管紧张素Ⅱ(angiotensinⅡ,AngⅡ)的含量;采用硝酸还原酶法测定血清中一氧化氮(nitric oxide,NO)含量,Griess化学比色法测定血清中一氧化氮合酶(nitric oxide sythase,NOS)活性的变化。结果:与模型组比较,大蓟组(高低剂量组)大鼠血压显著降低;而血清NO、NOS水平显著升高,血浆中AngⅡ的含量降低,差异均有显著性。结论:大蓟对肾性高血压有降压作用。
Objective: To observe the effect and mechanism of alcohol extract of Artichoke on 2-kidney 1-clip (2K1C) hypertensive rats. Methods: The 2K1C model of renal hypertensive rat was used to measure the changes of tail arterial pressure before and after the administration of alcoholic extract of Artichoke, and the levels of angiotensin Ⅱ (AngⅡ) in plasma were detected by radioimmunoassay. . The content of nitric oxide (NO) in serum was measured by nitrate reductase method, and the activity of nitric oxide synthase (NOS) in serum was measured by Griess colorimetry. Results: Compared with the model group, the blood pressure of the rats in the Ji-Ji group (high-low dose group) decreased significantly; while the levels of NO and NOS in serum increased significantly and the content of AngⅡ in the plasma decreased. The difference was significant. Conclusion: Thistle has antihypertensive effects on renal hypertension.
