以柠条为原料,利用HPLC技术研究了在超声波辅助常压处理下,提取时间、提取温度、乙醇提取液浓度及料液比对柠条叶中黄酮类化合物芦丁提取的影响。通过正交试验确定最佳提取参数,并建立芦丁的高效液相色谱(HPLC)法测定方法。结果表明:乙醇浓度和提取时间对芦丁提取量有显著影响,最佳提取参数为:60min,60℃,乙醇浓度80%,液料比1:10时,柠条叶中芦丁含量为101.84×10-2mg/g。
Using Caragana korshinskii as raw material, the effects of extraction time, extraction temperature, concentration of ethanol extract and solid-liquid ratio on the extraction of rutin from the leaves of Caragana korshinskii were studied by HPLC. The orthogonal test was used to determine the optimal extraction parameters, and the determination method of rutin was established by high performance liquid chromatography (HPLC). The results showed that ethanol concentration and extraction time had significant effects on the extraction of rutin. The optimal extraction parameters were as follows: 60 min, 60 ℃, ethanol concentration 80%, the ratio of liquid to solid 1:10, the content of rutin in the leaves was 101.84 × 10-2mg / g.
