利用组氨酸与锌离子的特异性结合,用水溶性CdSe/ZnS核壳型量子点标记朊蛋白(PrPC).研究了量子点标记朊蛋白的荧光光谱、琼脂糖凝胶电泳以及二者结合比.标记后荧光强度增加两倍多,二者结合比为3∶1.这种方法能快速简便地实现朊蛋白的定位点标记,为进一步深入研究朊蛋白提供了一种新的标记手段.
The prion protein (PrPC) was labeled with water-soluble CdSe / ZnS core-shell quantum dots using the specific binding of histidine to zinc ions.The fluorescence spectra of quantum dot-labeled prion protein, agarose gel electrophoresis, The fluorescence intensity of the labeled protein increased more than two times and the binding ratio of the two was 3:1.The method could quickly and easily achieve the site mark of prion protein and provided a new marker for further study of prion protein.
