在体外用雷公藤多甙(TⅡ)处理17例正常人及8例类风湿关节炎(RA)患者的外周血单个核细胞或淋巴细胞或单核细胞,不同组合的细胞在南陆素刺激下培养2周,用双夹心固相免疫放射法测定培养上清液中的IgG及IgM水平。结果发现TⅡ对正常人及RA患者PBMC体外分泌IgG及IgM的抑制作用相近,TⅡ单独处理单核细胞或/及淋巴细胞均可抑制Ig的分泌,但对后者的抑制作用明显强于前者。此结果与全制剂、青霉胺等抗风湿药有所不同。本实验结果与该药的临床资料相一致,提示TⅡ的治疗作用与它的免疫抑制作用相关。
Peripheral blood mononuclear cells or lymphocytes or mononuclear cells from 17 normal subjects and 8 rheumatoid arthritis (RA) patients were treated with tripterygium glycosides (TII) in vitro. Cells from different combinations were stimulated with stilbene After 2 weeks of culture, the IgG and IgM levels in the culture supernatant were measured by double-sandwich solid phase immunoradiometric assay. The results showed that TⅡinhibited the secretion of IgG and IgM in PBMCs in normal and RA patients in vitro. TⅡinhibit the secretion of Ig in monocytes and / or lymphocytes, but the inhibitory effect was stronger than the former. This result and the whole preparation, penicillamine and other antirheumatic drugs are different. The experimental results consistent with the clinical data of the drug, suggesting that the therapeutic effect of T Ⅱ and its immunosuppressive effect.
