目的 :研究紫杉醇对小鼠巨噬细胞免疫活性的调节作用。方法 :按常规方法收集纯化 8~ 12周龄BALB/c雌性小鼠腹腔巨噬细胞 ,用RPMI16 40培养基培养 ,按所用药物紫杉醇 (Paclitaxel)不同浓度和 (或 )干扰素 γ(INF γ)进行分组并设对照组。一氧化氮 (NO)测定采用Griess法。结果 :一定浓度的紫杉醇能够诱导巨噬细胞产生NO ,且随浓度增大而增加 ;紫杉醇联合INF γ时 ,诱导作用明显增加 ,且紫杉醇腹腔给药更易达到有效活化巨噬细胞浓度。结论 :腹腔给药有利于紫杉醇发挥抗肿瘤作用 ,若与INF γ联合更能发挥其免疫化学治疗作用
Objective : To study the regulatory effect of paclitaxel on immune activity of mouse macrophages. METHODS: Peritoneal macrophages of BALB/c female mice aged 8 to 12 weeks were collected and purified by conventional methods and cultured in RPMI1640 medium at different concentrations of paclitaxel (or IFN gamma (INF γ)). Group and set up a control group. Nitric oxide (NO) was measured using the Griess method. RESULTS: A certain concentration of paclitaxel could induce NO production in macrophages and increase with the increase of concentration. When paclitaxel was combined with INF gamma, the induction effect was significantly increased, and paclitaxel was easier to achieve effective activation of macrophage concentration after intraperitoneal administration. Conclusion : Intraperitoneal administration is beneficial to the anti-tumor effect of paclitaxel. If combined with INF γ, it can exert its immunochemotherapy effect.
