目的研究CCR6-CCL20对食管鳞癌患者体内Th17细胞分布的影响。方法采用流式细胞术检测食管鳞癌患者手术切除的癌组织、癌旁组织及外周血中Th17细胞的比例及CCR6+Th17细胞的比例。RT-PCR方法检测组织中CCL20 mRNA水平。结果与癌旁组织及外周血相比,癌组织中Th17细胞明显升高(P<0.01);癌组织中CCR6+Th17细胞的比例高于癌旁组织及外周血(P<0.01);癌组织中CCL20 mRNA水平高于癌旁组织(P<0.01),并与Th17呈正相关。结论 CCR6-CCL20趋化轴可能在食管鳞癌患者Th17细胞浸润到肿瘤组织中发挥重要作用。
Objective To investigate the effect of CCR6-CCL20 on the distribution of Th17 cells in esophageal squamous cell carcinoma patients. Methods Flow cytometry was used to detect the proportion of Th17 cells and the proportion of CCR6 + Th17 cells in surgically resected esophageal squamous cell carcinoma tissues, paracancerous tissues and peripheral blood. The level of CCL20 mRNA was detected by RT-PCR. Results Compared with adjacent tissues and peripheral blood, Th17 cells in cancer tissues were significantly increased (P <0.01). The proportion of CCR6 + Th17 cells in cancer tissues was higher than that in paracancerous tissues and peripheral blood (P <0.01) The level of CCL20 mRNA was higher than that in paracancerous tissues (P <0.01), and positively correlated with Th17. Conclusion The CCR6-CCL20 chemokine axis may play an important role in the invasion of Th17 cells into tumor tissues in patients with esophageal squamous cell carcinoma.
