[目的]研究苯并 (a)芘[benzo(a)pyrene ,BaP]对血管内皮细胞钙依赖型钾通道 (Ca2 + _dependentK+ channel,KCa)的影响及其意义。[方法]原代培养猪主动脉血管内皮细胞 ,不同浓度的BaP(0、0.1、0.5、1、5、10μmol/L)染毒24h,以全细胞膜片钳技术检测各组细胞KCa 的变化情况。[结果]随BaP浓度的逐渐升高 ,内皮细胞膜KCa 开放程度扩大。在膜电位为 +40mV时 ,对照组和BaP染毒组 (0.1、0.5、1、5、10μmol/L)的外向电流分别为 (350.12±53.23)、(372.35±54.56)、(765.25±86.55)、(897.44±79.33)、(1245.65±89.75)、(1810.23±65) pA ,其中BaP(0.5、1、5、10μmol/L)染毒组的外向电流值与对照组相比 ,差异有显著性(P<0.05)。[结论]BaP升高血管内皮细胞内Ca2 +浓度 ,可能引起KCa 开放程度扩大。
[Objective] To investigate the effect and significance of benzo (a) pyrene and BaP on Ca2 + -dependent K + channel (KCa) in vascular endothelial cells. [Method] Primary cultured porcine aortic vascular endothelial cells were exposed to different concentrations of BaP (0,0.1,0.5,1,5,10μmol / L) for 24h, and the changes of KCa in all groups were detected by whole-cell patch clamp technique . [Result] With the gradual increase of BaP concentration, the opening degree of endothelial cell membrane KCa increased. The outward currents of control group and BaP group were (350.12 ± 53.23), (372.35 ± 54.56) and (765.25 ± 86.55) at the membrane potential of +40 mV, respectively , (897.44 ± 79.33), (1245.65 ± 89.75) and (1810.23 ± 65) pA, respectively, of which the outward currents of BaP (0.5, 1, 5 and 10μmol / L) (P <0.05). [Conclusion] BaP increases the intracellular Ca2 + concentration in vascular endothelial cells, which may lead to the expansion of KCa.
