以淀粉微球和人IgG为原料,环氧氯丙烷为活化剂,戊二醛为加固交联剂,制备免疫淀粉微球。以交联率为指标进行反应条件优化,得到免疫淀粉微球的最佳合成条件:环氧氯丙烷浓度20%、戊二醛浓度0.5 mol/L、反应时间9 h。并用荧光标记二抗进行体外免疫检测,计算了抗体交联量与淀粉微球粒径的对应关系。利用荧光倒置显微镜对产物进行形态分析,证明人IgG与淀粉微球产生共价交联。
The starch microspheres and human IgG as raw materials, epichlorohydrin as activator, glutaraldehyde as reinforcing cross-linking agent, preparation of immune starch microspheres. The reaction conditions were optimized with the cross-linking rate as the index, and the optimal conditions for the preparation of the immunized starch microspheres were obtained: epichlorohydrin concentration of 20%, glutaraldehyde concentration of 0.5 mol / L and reaction time of 9 h. In vitro immunofluorescence was carried out with fluorescently labeled secondary antibody, and the corresponding relationship between antibody cross-linking amount and particle size of starch microspheres was calculated. Morphological analysis of the product using a fluorescence inverted microscope demonstrated that human IgG covalently crosslinked with the starch microspheres.
