目的:探索基因芯片在人脑胶质瘤基因表达研究中的应用。方法:通过RT-PCR反转录、同位素标记、基因芯片方法研究5例星型胶质瘤和4例正常脑组织mRNA的表达差异。结果:在 14 892个可表达顺序片断(expressionsequencetag,EST)中,对照与2个胶质瘤样品间有102个差异表达。对照与3级样品间共有39个显著差异表达基因,与4级样品间共有78个显著差异表达基因,3级和4级肿瘤有5个共同异常表达基因。结论:基因芯片发现了胶质瘤的异常基因表达谱。
Objective: To explore the application of gene microarray in the gene expression of human glioma. Methods: RT-PCR reverse transcription, isotope labeling, gene chip method in 5 cases of astrocytoma and 4 cases of normal brain tissue mRNA expression differences. RESULTS: Of 14 892 ESTs, there were 102 differential expression between control and two glioma samples. There were 39 significant differentially expressed genes in control and grade 3 samples, 78 significantly differentially expressed genes in grade 4 and 5 common in grade 3 and 4 tumors. Conclusion: Abnormal gene expression pattern of glioma was found in the gene chip.
