以反相高效液相色谱法（ＲＰ－ＲＰＬＣ）测定葛根总黄酮提取物及制剂中葛根素的含量。检测波长２５０ｎｍ，流动相为甲醇－水（２５：７５），进样２０μｌ采用外标法。当葛根素浓度在０．５－１２μｇ／ｍｌ时，浓度与峰面积呈良好线性关系，ｒ＝０．９９９１。回收率及ＲＳＤ分别为１００．９％，１．２８％。
RP-RPLC was used to determine the content of puerarin in Pueraria total flavonoids extracts and preparations. Detection wavelength of 250nm, the mobile phase of methanol - water (25:75), injection of 20μl using external standard method. When the puerarin concentration of 0.5-12μg / ml, the concentration and peak area showed a good linear relationship, r = 0.9991. The recovery rate and RSD were 100.9% and 1.28% respectively.
