目的:探讨脂联素(ADPN)及其脂联素受体2(AdipoR2)对非酒精性脂肪性肝炎(NASH)病理影响及化痰祛湿活血方干预作用。方法:采用高脂饲料联合四环素腹腔注射建立大鼠NASH模型。造模第2周采用化痰祛湿活血方高、中、低剂量和多烯磷脂酰胆碱(易善复)胶囊干预观察高脂模型。实验结束时,进行肝组织HE染色,ADPN、AdipoR2免疫组化染色,观察病理改变及ADPN、AdipoR2表达。结果:与对照组比较,化痰祛湿活血方高剂量能明显升高ADPN、AdipoR2蛋白表达(P<0.01),减轻肝脂肪变性及炎症程度。结论:化痰祛湿活血方可能通过上调ADPN、AdipoR2表达,减轻肝脏炎症,改善肝细胞脂肪沉积。
Objective: To investigate the effects of adiponectin (ADPN) and AdipoR2 on the pathological changes of non-alcoholic steatohepatitis (NASH) and the therapeutic effect of Huatan Qushi Huoxue Fang. Methods: NASH model was established by intraperitoneal injection of high-fat diet combined with tetracycline. In the second week of the model, high, middle and low doses of phlegm dampness and blood circulation prescription and polyene phosphatidylcholine (Yi Shan Fu) capsule were used to observe the hyperlipidemia model. At the end of the experiment, HE staining of liver tissue, ADPN and AdipoR2 immunohistochemical staining were performed to observe the pathological changes and ADPN, AdipoR2 expression. Results: Compared with the control group, the high dose of Huatan Qushi Huoxue Recipe could significantly increase the ADPN and AdipoR2 protein expression (P <0.01), and reduce the degree of hepatic steatosis and inflammation. Conclusion: Huatan Qushi Huoxue Prescription may improve hepatic fat deposition by up-regulating ADPN and AdipoR2 expression, relieving liver inflammation.
