Caspase是一组与凋亡密切相关的蛋白质家族,参与多种神经疾病的发病机制。在脑卒中、神经系统变性病、多发性硬化和癫疒间等疾病发病过程中,均可观察到caspase的活化。Caspase抑制剂能抑制caspase活性,阻断caspase介导的细胞凋亡途径,具有神经保护作用,有可能为这些疾病的治疗开辟新的途径。具有caspase抑制作用的米诺环素(minocycline ,美满霉素)治疗亨廷顿病和肌萎缩侧索硬化症的临床试验目前正在进行中。本文主要综述了caspase抑制剂的种类、作用原理及其在动物和临床方面的研究结果。
Caspase is a group of protein family closely related to apoptosis involved in the pathogenesis of a variety of neurological diseases. In stroke, neurological degenerative diseases, multiple sclerosis and epilepsy and other diseases in the pathogenesis, can be observed caspase activation. Caspase inhibitors can inhibit caspase activity, block the caspase-mediated apoptotic pathway, have neuroprotective effects and may open up new avenues for the treatment of these diseases. Clinical trials of minocycline (minocycline) with caspase inhibition to treat Huntington’s disease and amyotrophic lateral sclerosis are currently underway. This article reviews the types of caspase inhibitors, their principles of action and their findings in animals and clinics.
