以大肠杆菌HB101菌株为受体菌,研究了CaCl2浓度、转化时间及转化后的温育时间对柯萨奇B组病毒3型(CVB3)cDNA重组质粒pGP51B转化作用的影响。实验结果表明,CaCl2浓度和转化时间对转化率的影响极为显著,而转化后的温育时间对转化率的影响不明显。本实验证明,以75mmo1/LCaCl2制备感受态细胞,转化30分钟后直接涂布于选择培养基平板,选择转化子为最佳转化条件,可达到高效、快速转化的目的。
The Escherichia coli HB101 strain was used as the recipient bacteria to study the effects of CaCl2 concentration, transformation time and the incubation time after transformation on the transformation of recombinant plasmid pGP51B of Coxsackie virus B (CVB3). The experimental results showed that the effect of CaCl2 concentration and conversion time on the conversion rate was significant, but the conversion time was not significantly affected by the incubation time. The experiment proves that competent cells can be prepared with 75mmo1 / LCaCl2, coated directly on the selection medium plate after transformation for 30 minutes, and the transformant is selected as the optimal transformation condition, so that high efficient and rapid transformation can be achieved.
