目的:筛选蛞蝓有效提取物,并观察蛞蝓有效提取物EL-3对人肺癌A549细胞增殖的抑制作用。方法:体外培养人肺癌A549细胞和中国仓鼠肺上皮CHL细胞;MTT比色法测定细胞活性;软琼脂集落形成法检测细胞锚定非依赖性生长能力。结果:以不同提取方式获得的6种蛞蝓提取物对人肺癌A549细胞增殖活性具有抑制作用,其中以EL-3抑制作用最强,其IC50值为9.4μg/mL,可作为蛞蝓有效提取物;EL-3选择性抑制A549细胞活性,其选择指数是43.6;EL-3能有效抑制A549细胞锚定非依赖性生长,呈浓度依赖性。结论:EL-3具有选择性抑制A549细胞增殖作用。
OBJECTIVE: To screen the effective extracts of alfalfa and observe the inhibitory effect of alfalfa effective extract EL-3 on the proliferation of human lung cancer A549 cells. METHODS: Human lung cancer A549 cells and Chinese hamster lung epithelial CHL cells were cultured in vitro. Cell viability was determined by MTT colorimetric assay. Soft agar colony formation assay was used to determine the ability of cell anchorage-independent growth. RESULTS: The six extracts obtained by different extraction methods had inhibitory effect on the proliferation activity of human lung cancer A549 cells, of which EL-3 had the strongest inhibitory effect and its IC50 value was 9.4 μg/mL, which could be used as an effective extract of alfalfa; EL-3 selectively inhibited the activity of A549 cells, its selection index was 43.6; EL-3 could effectively inhibit anchorage-independent growth of A549 cells in a concentration-dependent manner. Conclusion: EL-3 can selectively inhibit the proliferation of A549 cells.
